摘要
目的:观察丙酮酸乙酯(EP)对缺血缺氧性脑损伤新生大鼠脑组织Bcl-2及Bax蛋白表达的影响,探讨EP在新生大鼠缺血缺氧性脑损伤脑组织中保护作用的可能机制。方法:90只Wistar新生大鼠,随机分为3组:A组为对照组,B组为模型组,C组为干预组。给动物吸入含有92%氮气和8%氧气混合气体建立新生大鼠缺氧缺血性脑损伤(HIBD)动物模型,免疫组化法观察大鼠海马区Bax、Bcl-2表达变化。结果:A组Bax、Bcl-2表达变化不明显;B组Bcl-2表达明显减少,与A组、C组比较差异有统计学意义(P<0.05);C组Bcl-2表达较B组明显增多(P<0.05)。B组Bax表达明显增多,与A组、C组比较差异有统计学意义(P<0.05),C组Bax表达较B组明显减少(P<0.05)。结论:丙酮酸乙酯对新生大鼠缺血缺氧性脑损伤脑组织有保护性作用,此作用可能与其调节Bcl-2和Bax表达水平有关。
Objective: To observe the effects of ethyl pyruvate on expressions of Bcl-2 and Bax in neonatal rats hypoxic-ischemic brain damage,and discuss ethyl pyruvate' s possible mechanism of protection in hypoxic-ischemic brain damage. Methods: Ninety 7-day-old Wistar rats were randomly divided into 3 groups(n=30,respectively): normal control group(n=30, A), HIBD model group(n=30, B) ,Ethyl pyruvate theraphy group(n=30,C), The models of HIBD were established in neonatal rats by inhaling the mixed gases of 8~/,, 02 and 92% N2, The Bax and Bcl-2 expression were assayed by the immunohistochemical staining. Results: The expression of Bax and Bcl-2 was not significantly changed in hippocampus in group A. the expression of Bcl-2 was significantly reduced in group B ,the expression of Bcl-2 in group C was increased significantly than group B(P〈0. 05). The expression of Bax expression was increased significantly in group B ,the expression of Bax expression in group C was significantly reduced than group B (P〈0.05).Conclusions: Ethyl pyruvate has a protective role in neonatal rats hypoxic-ischemic brain damage brain tissue, this effect may be related to regulate the expression of Bcl-2 and Bax.
