摘要
目的:研究乳腺癌中DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶(MGMT)的表达与癌细胞对紫杉醇联合阿霉素(TA方案)敏感率的相关性。方法:采用细胞原代培养和MTT比色法观察TA方案对乳腺癌原代细胞培养的抑制率,运用SABC免疫组化法检测MGMT的基因表达情况。结果:在乳腺癌细胞中MGMT阳性率为55.42%(46/83),有淋巴结转移组MGMT阳性率明显高于无淋巴结转移组(P<0.05),TA方案敏感率为42.17%(35/83),MGMT表达与TA方案抑制率低度相关(r=0.23)。结论:TA方案的抑制率与MGMT的表达低度相关。
OBJECTIVE:To evaluate the relativity between the expression of O6-methylguanine-DNA methyltransferase(MGMT) and the sensitivity of breast carcinoma cell to paclitaxel combined with doxorubicin(TA scheme).METHODS:The inhibition rate of TA scheme on primary culture of breast carcinoma cell was observed by primary culture and MTT method,and the expression of MGMT was detected by SABC immunohistochemical method.RESULTS:In the breast carcinoma cells,the positive rate of MGMT was 55.42%(46/83).The positive rate of MGMT in lymphmetastasis group was higher than in non-lymphmetastasi significantly(P0.05),and the sensitive rate of TA scheme was 42.17%(35/83).There was low correlation between the expression of MGMT and the inhibition rate of TA scheme(r=0.23).CONCLUSION:The inhibition rate of TA scheme is correlated to the expression of MGMT.
