摘要
运用毛细管区带电泳(CZE),以pH=6.59的60mmol/L Na2HPO4-NaH2PO4作为电泳缓冲溶液,测定波长为200nm,还原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG)于5min内达到基线分离。GSH和GSSG的线性范围分别为4~1 000μmol/L和2~1 000μmol/L,相关系数R2值均为0.9999,迁移时间和峰面积的相对标准偏差分别小于0.58%和2.30%,GSH和GSSG的检出限分别为1.66μmol/L和0.86μmol/L,回收率在95.4%~103.3%之间,相对标准偏差均小于3.05%(n=3)。建立的新方法用于市售注射用还原型谷胱甘肽钠的分析,测得注射用还原型谷胱甘肽钠中GSSG含量为0.47%,光学纯度为99.53%。
A technique for the analysis of reduced glutathione sodium for injection by capillary zone electrophoresis(CZE) with diode array detector(DAD) has been reported.Separation of GSH and GSSG was achieved in 5 min using a 60 mmol/L Na2HPO4-NaH2PO4 buffer(pH=6.59).Linear range of GSH and GSSG were 4~1 000 μmol/L and 2~1 000 μmol/L,and relative standard deviation(RSD) of migration time and peak area were less than 0.58% and 2.30%,respectively.The detection limits were 1.66 mmol/L for GSH and 0.86 mmol/L for GSSG.The recoveries were in the range of 95.4%~103.3%,and the RSD for three independent analysis of each sample was less than 3.05%.The new method has been applied to the analysis of reduced glutathione sodium for injection,and the determination results were 0.47% for GSSG and 99.53% for GSH in reduced glutathione sodium for injection.
