尿中N,N-二甲基乙酰胺及其代谢物N-甲基乙酰胺的气相色谱测定法 被引量:7

摘要 目的:建立职业工人尿样中的N,N-二甲基乙酰胺(DMAC)及其代谢产物N-甲基乙酰胺(NMAC)的气相色谱测定方法。方法:尿样经冻融后离心,上清液加甲醇(1∶1,v/v)后用HP-INNOWAX毛细管柱分离,氢火焰离子化检测器(FID)对尿样进行检测,外标法定量。结果:DMAC和NMAC在0μg/ml~300.0μg/ml内呈线性关系;相关系数为0.9999;最低检出限分别为1.65μg/ml和2.90μg/ml;样品加标回收率分别为99.8%~100.4%和98.0%~99.9%。结论:该法操作简便、快速、准确、灵敏度高,适用于职业工人尿样中DMAC、NMAC的同时测定。 Objective:To develop a method to determine N,N-Dimethylacetamide(DMAC) and it′s metabolite N-Methylacetamide(NMAC) in worker′s urine by gas chromatography(GC).Methods: The urine was separated with Innowax capillary column and detected by Flame Ionization Detector(FID) after being extracted with methanol(1∶ 1,v/v).The qualification was accomplished by external standard method.Results: The linear ranges of DMAC and NMAC were 0 μg/ml to 300.0 μg/ml with correlation coefficient of both 0.9999.The detection limits,recovery ranges for DMAC and NMAC were 1.65 μg/ml and 2.90 μg/ml,99.8% to 100.4% and 98.0% to 99.9%,respectively.Conclusion: This method was simple,quick,accurate and sensitive.It could be applied to determine DMAC and NMAC in urine simultaneously.
