摘要
目的:建立尿中草酸和柠檬酸的离子排阻色谱测定方法。方法:采用紫外检测器,利用Rezex ROA有机酸分析色谱柱,以0.010 mol/L硫酸溶液为淋洗液,流速0.40 ml/min,采用紫外波长201 nm进行检测,外标法定量。结果:草酸和柠檬酸在相应的试验浓度范围内均具有良好的线性关系,其相关系数大于0.9990,草酸、柠檬酸检出限分别为0.2 mg/L、1.0 mg/L,平均回收率在88.3%~102.6%之间,RSD均小于5.0%。结论:该法具有简单、准确、灵敏等优点,适合尿中草酸和柠檬酸含量的检测。
Objective:To establish an analytical method for the determination of oxalic acid and citric acid in urine by ionic exclusion chromatography.Methods: The oxalic acid and citric acid in urine were separated on Rezex ROA organic acid column(300 mm×7.8 mm i.d.,8 μm) by using 0.010 mol/L sulfuric acid as an eluent at a flow rate of 0.40 ml/min.Detection was performed on an ultraviolet detector at λ=201 nm and quantification was calculated by an external standard method.Results: The linearity of oxalic acid and citric acid in the tested concentration range was good and the correlation coefficients were both more than 0.9990.The limits of quantification were found to be 0.2 mg/L and 1.0 mg/L for oxalic acid and citric acid.The extraction recoveries were between 88.3%~102.6% and the RSD were both less than 5.0%.Conclusion: This method is simple,sensitive and accurate for the determination of oxalic acid and citric acid in urine and it can be satisfactorily applied to the routine analysis.
