摘要
目的研究3'-大豆苷元磺酸钠对大鼠脑缺血/再灌注损伤的保护作用,并初步探讨其保护机制。方法大鼠大脑中动脉栓塞,制作脑缺血再灌注损伤模型,缺血后10 min,于舌下静脉给予不同剂量的3'-大豆苷元磺酸钠。再灌注24 h后,断头取脑,做TTC染色,测定脑梗死体积,另一组取血,离心后,取血清,测定血清一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)的活性及一氧化氮(NO),丙二醛(MDA)的含量。结果与模型组相比,各剂量的3′-大豆苷元磺酸钠治疗组脑梗塞体积减小;3'-大豆苷元磺酸钠治疗组血清NOS和SOD的活性升高,NO含量升高,MDA含量降低。结论 3'-大豆苷元磺酸钠对大鼠脑缺血再灌注损伤具有一定的保护作用,其保护机制与3'-大豆苷元磺酸钠促进NO释放及增强氧自由基的清除有关。
Objective To elucidate the protective effects of 3'-daidzein sulfonate sodium(DSS) on the ischemia/reperfusison injury in rats and its primary mechanism.Methods A rat focal cerebral ischemia/reperfusion injury model was established by using the intraluminal suture occlusion followed by reperfusion method.After occlusion for 10 minutes,3'-daidzein sulfonate sodium at the indicated doses was given by sublingual vein injection.24 h after reperfusion,the cerebral tissue was stained with the TTC method,and the infarction volume was investigated.In another group,blood samples were collected and serum NOS,SOD activities and NO,MDA contents were decected.Results Compared with model group,3'-daidzein sulfonate sodium could decrease brain infarction colume and serum MDA content,elecate serum NOS and SOD activities and the NO content.Conclusion 3'-daidzein has protective effect on cerebral ischemia reperfusion injury.The mechanism may be related with increase NO release and enhacement of oxygen free radical elimination.
