醛固酮对h9c2心肌细胞缝隙连接蛋白43的影响 被引量:1

摘要 目的:观察醛固酮对h9c2心肌细胞缝隙连接蛋白43(Connexin 43,Cx43)表达的影响。方法:将h9c2心肌细胞随机分为对照组和实验组,实验组用不同浓度(1×10-8、1×10-6、1×10-4 mol/L)醛固酮处理24 h后,用RT-PCR和Western Blot观察不同浓度醛固酮下h9c2心肌细胞Cx43蛋白和mRNA的变化。结果:1×10-8 mol/L醛固酮组Cx43mRNA和蛋白表达含量较对照组增加;1×10-6 mol/L和1×10-4 mol/L组的Cx43蛋白表达含量较对照组减少,而Cx43mRNA较对照组无明显改变。结论:醛固酮可能是通过对表达Cx43的双重影响来参与心肌细胞间隙连接重构的。 Objective:To investigate the effects of aldosterone on the expression of Cx43 in h9c2 cells.Methods:The h9c2 cells were randomly divided into control group and experimental group.The experimental group was exposed to various concentration of aldosterone(1×10^-8,1×10^-6,1×10^-4mol/L) for 24 hours,the changes of Cx43 protein and Cx43 mRNA were investigated by Western Blot and RT-PCR.Results:Compared with the control group,the expression of Cx43 protein and Cx43 mRNA increased in 1×10^-8mol/L aldosterone group,but the Cx43 protein decreased in 1×10^-6 mol/L and 1×10^-4 mol/L aldosterone group,the Cx43 mRNA was no obvious difference.Conclusion:Aldosterone may be involved in gap junction remodeling through its dual effects on the expression of Cx43.
