摘要
利用ISSR分子标记,以云南木犀[Osmanthus yunnanensis(Franchet)P.S.Green]和柊树[O.heterophyllus(G.Don.)P.S.Green]为对照种,对6个省(市)45份古桂花和2份野生桂花进行了遗传多样性研究。结果表明,古桂花具有丰富的遗传多样性,选用的14条引物共扩增出148条DNA条带,其中多态性条带131条,占88.51%;平均观察等位基因数、有效等位基因数、Nei's遗传多样性指数和Shannon多样性信息指数分别为1.885 1、1.410 6、0.241 5和0.368 8;聚类分析结果表明,对照种与桂花遗传距离较远,最早分开;古桂花具有很强的地理相关性,来自同一来源地或具有相似颜色的个体往往聚在一起;四季桂与其他秋桂类个体遗传距离较远。
Interspecific relationships of 45 ancient sweet osmanthus trees from 6 provinces and 2 wild species of sweet osmanthus were evaluated by using ISSR(inter simple sequence repeat) marker with Osmanthus yunnanensis(Franchet) P.S.Green and O.fordii Hemsl as contrast species.The results showed that ancient sweet osmanthus trees had abundant genetic diversity.Fourteen ISSR primers produced a total of 148 bands,of which 131 were polymorphic.The percentage of polymorphic bands was 88.51%.The mean observed number of alleles,effective number of alleles,Nei′s genetic distance and Shannon index were 1.885 1,1.410 6,0.241 5 and 0.368 8 respectively.The cluster analysis showed that the genetic distance between contrast species and sweet osmanthus was far.Ancient sweet osmanthus trees were closely correlated with geographical locations,and trees from the same place or in similar color tended to cluster together.The genetic distance between asiaticus group and others was far.
