摘要
利用反相高效液相色谱法测定西乔3号中芦丁、槲皮素中成分的含量。采用SHIMDZU VP-ODS(4.6 mm×150 mm,5.0μm)色谱柱,甲醇-0.08%:磷酸溶液(6∶4)为流动相,流量为0.95 mL.min-1,检测波长为358 nm。待测组分与其他组分分离度良好(R>1.5),芦丁和槲皮素线性范围分别为32.0~288.0μg.ml-1和18.8~169.26μg.ml-1,平均回收率分别为98.5%,99.0%,相对平均偏差(n=5)分别为0.4%,0.4%。
This paper is to introduce the experiment of determining the contents of rutin and quercetin in Tartary Buckwheat No.3 with RP-HPLC.Taking SHIMDZU VP-ODS column(4.6 mm×150 mm,5.0 μm),a mixture of methanol-0.08% phosphoric acid in the ratio of 6 to 4 by volume with a flow-rate of 0.95 mL·min-1 and wavelength of 358 nm as the mobile phase,the range of linearity of rutin and quercetin(values of peak area and mass concentration),after being well-separated,is between 32~288 μg·ml-1 and 18.2~169.2 μg·ml-1 respectively,the average rate of recovery for rutin is 98.5%,and 99.0% for quercetin with values of RSD's(n = 5) 0.4% and 0.4% respetively.
