胭脂鱼细胞色素b基因序列测定及亚口鱼科系统进化分析 被引量:1

Sequence of mitochondrial cytochrome b gene of Myxocyprinus asiaticus and phylogeny of Catostomidae fishes

摘要 为了解胭脂鱼(Myxocyprinus asiaticus)的遗传变异及亚口鱼(Catostomidae fish)的分子系统学,测定了3尾胭脂鱼的细胞色素b基因序列,并结合GenBank上5条亚口鱼科鱼类的相应序列一并分析。结果如下:1)共获得3尾胭脂鱼的细胞色素b基因1144 bp的一致序列,三条序列完全相同,无变异位点。2)构建的系统发育树显示,颈棱亚口鱼属(Xyrauchen texanus)和吸口鱼属(Moxostoma robustum)构成1支,胭脂鱼属(M yxocyprinus)、长背亚口鱼属(Cycleptus elongatus)和牛胭脂鱼属(Ictiobus)构成另1分支。3)自然选择检验显示,该基因在进化过程中主要受到负选择的作用。4)胭脂鱼属和牛胭脂鱼属的分歧时间最短(大约3.725百万年),颈棱亚口鱼属和长背亚口鱼属的分歧时间最长(大约5.375百万年)。 In order to study genetic variation of Myxocyprinus asiaticus and phylogeny of family Catostomidae,the mitochondrial cytochrome b(mt DNA cytb) gene of three individuals of M.asiaticus were sequenced,and analysed with five mt DNA cytb gene sequences of Catostomidae which downloaded from GenBank.The results were as follows:1) 1144 base pairs of consensus sequence was obtained from three M.asiaticus individuals,and these three sequences were identical without variation positions.2) Phylogenetic tree showed that genus Xyrauchen and Moxostoma formed one monophyletic clade,and genus Myxocyprinus,Cycleptus and Ictiobus formed another clade.3) mtDNA cytb gene of Catostomidae fishes was mainly acted by purifying selection.4) The divergence time between genus Myxocyprinus and genus Ictiobus was the shortest about 3.725 million years.And the divergence time between genus Xyrauchen and Cycleptus was the longest about 5.375 million years.
