异氟烷和七氟烷对老年小鼠认知功能的影响及其机制研究 被引量:8

摘要 目的探讨异氟烷和七氟烷对老年小鼠认知功能的影响及与胆碱能系统的关系。方法健康雌性18个月龄C57BL/6J老年小鼠30只,分为3组,每组10只。1.2%异氟烷(异氟烷组)或2.6%七氟烷(七氟烷组)麻醉6h(同时吸入30%氧气和70%氮气混合气体),未麻醉组为对照组。麻醉后2周行Morris水迷宫实验检测认知功能。水迷宫实验结束后取脑组织测定乙酰胆碱酯酶(AchE)和乙酰胆碱转移酶(ChAT)的活性和含量。结果与对照组比较,异氟烷组潜伏期和游泳路程均延长(P<0.05),小鼠脑组织内ChAT含量降低;七氟烷组潜伏期和游泳路程均无显著变化(P>0.05),AchE和ChAT的活性及含量均无显著变化。3组之间目标象限的时间和路程百分比均无显著性差异(P>0.05)。结论异氟烷麻醉后老年小鼠出现术后认知功能障碍(POCD),可能与ChAT含量减少有关。七氟烷麻醉后老年小鼠未出现POCD,且AchE和ChAT的活性及含量均无变化。 Objective To investigate the effects of isoflurane and sevoflurane on cognitive performance in aged mice and the mechanisms involved in cholinergic system.Methods Thirty female C57BL/6J mice(aged 18 months) were divided into three groups and anaesthetized with 1.2% isoflurane(n=10) or 2.6% sevoflurane(n=10) in oxygen/nitrogen(FiO2=30%) for 6 h,non-anaesthetized mice served as controls(n=10).Two weeks later,neurocognitive functions were evaluated by Morris water maze.After Morris water maze test,activation and content of the acetylcholine esterase(AchE) and choline acetyltransferase(ChAT) in the brain were evaluated.Results Compared with the control group,both latency and pathway of isoflurane anaesthetized group was increased(P 0.05),while the content of ChAT was decreased(P 0.05);neither latency nor pathway of sevoflurane group was different from those of the controls(P 0.05).The same situation was also observed in the activation and content of AchE and ChAT(P 0.05).One-factor analysis of variance showed that the percentage time and distance spent in the target quadrant had no significant difference among the three groups.Conclusions Isoflurane induced POCD of aged mice maybe induced by decrease of the content of ChAT.Aged mice have not shown POCD after sevoflurane anaesthesia.Neither the activation nor the content of AchE and ChAT is changed.
