MMP12和TIMP2 mRNA在大鼠舌黏膜癌变过程中的表达 被引量:1

摘要 目的观察基质金属蛋白酶-12(MMP12)及基质金属蛋白酶组织抑制因子-2(TIMP2)mRNA在4-亚基硝氧喹啉(4NQO)诱发大鼠舌黏膜癌变过程中的动态变化,探讨其在实验性大鼠舌黏膜癌变中的作用。方法 60只雌性SD大鼠随机分为3组,分别给予正常饮水、25 ppm(mg/L)的4NQO水溶液16、24周。舌部病变组织分成两等分,分别进行组织病理学分析和荧光定量PCR检测。结果 (1)4NQO诱发大鼠舌黏膜癌变过程中,正常饮水组无组织病理学改变,4NQO诱癌大鼠16周舌黏膜出现上皮中-重度异常增生,诱癌24周后主要病理学变化为舌鳞状细胞癌。(2)与正常组织相比,MMP12 mRNA表达水平在癌前病变中升高1.51倍,在舌癌中升高4.86倍(P<0.01),MMP12 mRNA表达随病理程度增加呈现递增趋势;与正常组织相比,TIMP2 mRNA在癌前病变中表达升高2.62倍(P<0.01),在舌癌中表达升高2.06倍(P<0.05),TIMP2 mRNA表达随病理程度增加呈现先增加后下降的趋势;舌黏膜癌变过程中MMP12与TIMP2 mRNA表达比例失调。结论 MMP12、TIMP2 mRNA的高表达参与4NQO诱发的大鼠舌黏膜癌变过程,两者表达的比例失调与舌黏膜癌变密切相关。
