摘要
该研究建立了高效液相色谱法定量测定烟丝及烟气中的单糖、双糖,确定的色谱条件如下:Waters NH2氨基色谱柱(250 mm×4.6 mm),乙腈︰水的体积比85︰15为流动相,流速1.0 mL/min,示差折光检测器,40℃柱温,进样量10μL,外标法定量。该方法的精密度RSD<5%,回收率大于96%。在此色谱条件下20 min内完成了单糖(葡萄糖、果糖、半乳糖)、双糖(蔗糖、麦芽糖)的分离,结果比较理想。研究表明此新建方法准确而快速,并能适用于烟丝及烟气中单糖、双糖的测定。
The method of HPLC was developed for determining the carbohydrates and monosaccharides of tobacco and smoke by using Waters HPLC with 250 mm×4.6 mm NH2 column and dual detectors.The mobile phase was composed of acetonitrile and water(85∶15),at a flow rate of 1.0 mL/min.The added quantity is 6 mL.The temperature of the column was 40 ℃.The method was quantified by an external standard mode with lower RSD than 5% and greater recovery than 96%.Under these conditions,the arbohydrates and monosaccharides were determined in 20 min with satisfactory results.This new method is rapid,simple and accurate,and useful for determining the carbohydrates and monosaccharides.
