摘要
目的将纳米羟磷灰石/壳聚糖(nHA/CS)支架材料与大鼠的成骨细胞在体外复合培养,研究其生物相容性,以期在骨组织工程支架材料的选择方面提供依据。方法原代培养大鼠成骨细胞,倒置显微镜作细胞形态学观察,碱性磷酸酶(ALP)染色和茜素红钙染色对细胞进行鉴定。将第三代细胞与nHA/CS材料体外复合培养,采用MTT比色法和生长曲线描绘测定支架材料对细胞增殖的影响。用SPSS13.0软件包对测定结果进行单因素方差分析。结果大鼠成骨细胞能在nHA/CS材料上附着、增殖,其生长良好,生理功能不受抑制。结论纳米羟基磷灰石/壳聚糖复合多孔支架材料与成骨细胞具有良好的生物相容性,可作为骨组织工程的理想支架材料之一。
Aim To investigate the feasibility of nHA/CS as scaffold material in bone tissue engineering.Methods The rat osteoblasts were isolated and cultured,then co-cultured with nHA/CS in vitro.The cells were observed under inverted microscope,cytochemical staining for ALP and ARS,MTT and growth curve were used for detection of cell proliferation.SPSS 13.0 software package was used for one-way ANOVA.Results Osteoblast could attach and extend on nHA/CS,whose proliferation and growth were not affected by nHA/CS.Conclusion nHA/CS is a good scaffold material for the bone tissue engineering.
