β-淀粉样肽对原代培养小鼠大脑皮质小胶质细胞形态学及细胞因子表达的影响 被引量:3

Effect of β-amyloid on the Morphology and Cytokine Production of Primary Cultured Mice Cortical Microglia

摘要 目的:研究原代培养的BALB/c小鼠大脑皮质小胶质细胞经β-淀粉样肽1-42(Aβ1-42)刺激后的形态学变化及细胞因子表达情况。方法:小胶质细胞用2次温和胰酶消化法分离,经预处理的Aβ1-42刺激后,相差显微镜观察形态学改变,RT-PCR检测IL-1β、-6基因表达水平。结果:小胶质细胞经Aβ1-42刺激后6 h细胞形态学发生明显改变;刺激24 h后IL-1β、-6基因表达水平明显上调(P<0.05)。结论:Aβ1-42可明显活化小胶质细胞,并诱导炎性细胞因子的表达。 Objective:To investigate the effect of β-amyloid 1-42(Aβ1-42)on the morphology and cytokine production of primary cultured BALB/c mice cortical microglia.Methods:The microglia were isolated by two-step mild trypsin method.After treatment with Aβ1-42,the morphology of microglia was observed under phase-contrast microscopy,and the gene expressions of IL-1β,-6 were detected.Results:After treatment with Aβ1-42 for 6 hours,the microglia showed a typical activated morphology.After 24 hours,the gene expressions of IL-1β,-6 were significantly higher than control(P〈0.05).Conclusion:The microglia can be activated and can express high level IL-1β,-6 after it is stimulated by Aβ1-42.
