摘要
为了研究谷胱甘肽过氧化酶家族(GPxs)在山羊不同组织表达的特异性及在睾丸不同发育时期的表达特性。采用SYBR Green荧光定量PCR的方法对太行青山羊公羊各组织和不同发育时期睾丸GPxsmRNA的表达进行了定量分析。结果表明:GPx1、GPx3、GPx4和GPx7在各组织广泛表达,其表达量由高到低的顺序:GPx1是肝脏>肺脏>心脏>睾丸、脾脏>肾脏>背最长肌,肝脏GPx1mRNA表达量是肌肉组织的6.7倍;GPx3是肾脏>心脏>肝脏、睾丸>脾脏、肺脏、背最长肌,肾脏GPx3mRNA表达量分别是肝脏和肺脏表达量的12和24倍;GPx4是睾丸>心脏>肾脏、肝脏、肺脏>脾脏、背最长肌,睾丸GPx4mRNA表达量分别是肝脏和肌肉的21和137倍;GPx7是肝脏>脾脏、肺脏、背最长肌>心脏>肾脏、睾丸。GPx5和GPx6仅在睾丸和附睾中表达,且GPx5在附睾头中的表达是睾丸的114倍;睾丸中GPx3表达量随日龄增加而降低,GPx1和GPx4表达量随日龄增加而增加,GPx5在90d开始表达且直接到达高峰平台,GPx6和GPx7在60d开始表达,90d到高峰平台,而后其表达量降低。山羊GPxs mRNA表达具有明显的组织特异性,山羊睾丸中GPxs mRNA表达具有时间依赖性。
The objectives of this study were to evaluate the tissue-specific expression of GPxs in various tissues of the male goat and investigate the expression of GPxs in testes at different stages of male goats.Total RNA was isolated from various tissues,and mRNA expression was detected by real-time fluorescent quantitative PCR.The quantitative real-time PCR results showed that the order of GPx1 mRNA expression level was liver lung heart testes,spleen kidney Longissimus dorsi(LD),the expression of GPx1 in liver was 6.7 times higher than that in LD;The order of GPx3 mRNA expression level was kidney heart liver,testes spleen,lung and LD,the expression of GPx3 in kidney was respectively 12 and 24 times higher than that in liver and lung,respectively;The order of GPx4 mRNA expression level was testes heart kidney,liver and lung spleen and LD,the expression of GPx4 in testes was respectively 21 and 137 times higher than that in liver and LD;The GPx5 and GPx6 mRNA only were expressed in testes and epididymidis.The expression of GPx5 mRNA in epididymidis head was 114 times higher than that in testes;The GPx7 mRNA appeared to be ubiquitously expressed almost in all tissues,the rank order of GPx7 mRNA expression was liverspleen,lung and LD heart kidney,testes.The expression of GPxs in goat testes was affected by age.The GPx3 mRNA expression decreased with the increase of age;The GPx1 and GPx4 mRNA expression developmentally increased as goats became older;The GPx5 mRNA was expressed and directly reached highest level at the age of 90 days;The GPx6 and GPx7 mRNA were expressed at the age of 60 days,and also reached the highest levels at the age of 90 days,then decreased at the age of 120 and 360 days.The expression of goat GPxs mRNA is tissue-specific pattern in various tissues,and the expression of GPxs mRNA is age-related pattern in goat testes.
