摘要
采用组织分离法从稻曲球上分离到病原真菌,通过柯赫法则鉴定,该病原菌为稻曲病菌。对稻曲病菌的实验室培养条件进行优化,结果表明,该菌能够在PDA培养基上正常生长,其生长温度在15~35℃,最适生长温度为28℃,温度过低或过高均抑制菌丝生长;在pH为4~10的PDA培养基上均能生长,最适pH值为5~8;同时测定了不同碳源和氮源对菌丝生长的影响,其中蔗糖和硝酸钙对菌丝生长最为有利;病原菌在胁本哲氏培养基中生长最快。
The pathogen was isolated from diseased plant.Through Koch's postulates,the pathogen was identified as Ustilaginoidea virens(Cooke)Tak.The study on culture conditions of the fungus in laboratory showed that the tested strain grew normally on PDA culture medium.The temperature range for its growth was between 15℃ to 35℃ and the optimal growth temperature was 28℃.The low or high temperature completely inhibited the mycelium growth.The range of pH was 4-10,and the optimal pH was 5-8.Sucrose and Ca(NO3)2 were separately the best carbon and nitrogen source for the mycelium growth among the tested carbon sources.The pathogen grew the fastest on Wakimoto toceshi medium among the solid culture media tested.
