摘要
通过单因素试验和"二次回归正交旋转组合设计"方法,在液体培养条件下,研究了不同发酵条件对1株野生紫芝菌丝体产量的影响。建立了装液量、初始pH、转速、培养温度和接种量5个因素的正交回归模型:Y=2.85701+0.17458X1-0.19802X12-0.20552X22-0.19302X32-0.15802X42-0.12302X52。从模型推知,装液量90 mL,pH6.0,转速140 r/m,培养温度27℃,接种量10.5%时,紫芝菌丝达到最大生物量3.15 g,验证实验结果与模型值相符。
In this paper,with liquid fermentation,both Single Factor Test and the Quadratic orthogonal Regressive Rotational Combination Test method were used to analyze the effect of different fermentation conditions on the mycelium biomass of one wild Ganoderma sinensis strain.The quadratic orthogonal regression model is of mycelium biomass(Y) to five factors of liquid volume(X1),initial pH value(X2),rotation speed(X3),culture temperature(X4),vaccinate amount(X5) and established as Y=2.85701+0.17458X1-0.19802X21-0.20552X22-0.19302X23-0.15802 X24-0.12302 X25.It was concluded from the model that when liquid volume,initial pH value,rotation speed,culture temperature,vaccinate amount were 90 mL,6.0,140 r/min,27 ℃ and 10.5 % respectively,the mycelium biomass(Y) was the maximal(3.15 g) and consistent with the confirmatory experiment result.
