摘要
为研究海带的高温适应机制,用不同的温度(15,20,25,28,30℃)处理海带配子体,观察记录配子体的生长状态,并测定其叶绿素a、脯氨酸、可溶性糖、丙二醛的含量以及POD活性随培养时间(1,3,5,7d)的变化趋势。试验结果表明,在20℃条件下,海带配子体的生长和各项生理指标与对照组15℃相比无明显变化;在25℃条件下培养1d后,除叶绿素a含量逐渐下降外,其余各项生理指标均出现上升的趋势;在28,30℃条件下,培养1d后,叶绿素a含量明显下降,细胞内容物萎缩分离,颜色变浅,3d后细胞内容物进一步萎缩,细胞膜破裂,5d后细胞大量死亡。
Some physiological indexes including the content of chlorophyll a,proline,soluble sugar,MDA and the activity of POD of the gametophytes of Laminaria japonica were measured under different temperatures(15,20,25,28,30 ℃) and different times(1,3,5,7 d).The results showed that all of the measured contents and activity of gametophytes of Laminaria japonica had little change under 20 ℃.When the gametophytes were cultured 1 day at 25 ℃,the contents of the physiological indexes ascended except for chlorophyll a.When the gametophytes were cultured 1 day under 28 ℃ and 30 ℃,the content of chlorophyll a descended obviously,the cellular content shrunk and separated,the pigment of the cell faded,the cytomembrane was ruptured after 3 days,huge amounts of cells died after 5 days.
