摘要
目的了解辣椒素对人结肠癌SW480细胞株增殖作用的影响,并探讨其可能的作用机制。方法采用MTT检测法,观察终浓度100-400μmol/L的辣椒素作用24、48 h对人结肠癌SW480细胞株生长作用的影响;采用流式细胞仪检测400μmol/L辣椒素作用人结肠癌SW480细胞株后其细胞周期变化和凋亡率。结果辣椒素能抑制结肠癌SW480细胞株增殖,呈浓度、时间依赖性。在400μmol/L辣椒素作用下,结肠癌SW480细胞株停滞于G0/G1期。辣椒素作用24、48 h,SW480细胞凋亡率升高(F=8.73、8.10,q=4.86-7.30,P〈0.01)。结论辣椒素抑制SW480细胞增殖,并诱导其凋亡,其机制可能是阻滞细胞周期于G0/G1期。
Objective To study the effect of capsaicin on proliferation of human colon-carcinoma cell line(SW480) and its possible mechanism. Methods MTT colorimetry was used to check the effect of 24-,48-hour action of capsaicin on proliferation of SW480 with dosage ranged from 100-400 μmol/L.The cell cycle and apoptosis under the capsaicin with a dosage of 400 μmol/L was detected by flow cytometry. Results Capsaicin inhibited the proliferation of SW480 cell line in a dosage-and time-dependent manner.At the dosage of 400 μmol/L,SW480 stagnated at phase G0/G1.The apoptosis rate increased after capsaicin action for 24 and 48 hours compared with that of the control group(F=8.73,8.10;q=4.86-7.30;P〈0.01). Conclusion Capsaicin in vitro inhibits proliferation of SW480 cells and induces their apoptosis,blockage of cell cycle at phase G0/G1 might be its mechanism.
