摘要
目的：通过体外实验探讨紫杉醇对恶性胶质瘤９Ｌ细胞株有否凋亡诱导作用。方法：在经ＭＴＴ比色法测试到紫杉醇对９Ｌ细胞具有细胞毒作用的前提下，进一步通过形态学观察，ＤＮＡ琼脂糖凝胶电泳及流式细胞仪来检测其细胞毒作用是否与诱导细胞凋亡有关。结果：紫杉醇对９Ｌ细胞具显著细胞毒作用；０１μｍｏｌ／Ｌ紫杉醇作用下９Ｌ细胞分裂主要被阻滞在Ｇ２／Ｍ期，并在第７２ｈ出现凋亡细胞峰，此时培养细胞ＨＥ染色表现出典型的凋亡细胞形态特征，抽提其ＤＮＡ做琼脂糖凝胶电泳显示凋亡特有的ＤＮＡ“阶梯”。结论：紫杉醇具有抗胶质瘤作用，其机制与诱导细胞凋亡有关，通过进一步的体内实验，可为其最终广泛应用于临床脑胶质瘤病人提供充分的理论依据。
Objective:The subject studied whether taxol can induce apoptosis in malignant glioma cells in vitro in order to offer sufficient basis for the treatment of malignant glioma.Methods:Malignant glioma cell line 9L were treated with different amount of taxol.MTT colorimetric assay was first employed to investigate cytotoxic effect of taxol on 9L cells.Then,morphology in microscopy,flow cytometry and DNA agerose gel electrophoresis were used to indicate apoptosis of 9L cells induced by taxol.Results:Taxol had obvious cytotoxic effect on 9L cells that is dose-dependent.0.1mol/L taxol treated 9L cells were arrested mainly in G2/M phase appeared the morphologic changes of apoptosis,DNA ladder and apoptosis peak.Conclusion:Taxol is effective in growth inhibition of malignant glioma,which is related to apoptosis inducement.
