摘要
观察猪传染性胃肠炎病毒对猪小肠黏膜上皮细胞Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶活性,细胞增殖和凋亡的影响。将猪传染性胃肠炎病毒接种于体外培养猪小肠黏膜上皮细胞24、48和72 h后,检测细胞的ATP酶(Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶)活性,MTT法检测细胞增殖能力,流式细胞仪测定细胞周期,PI染色法检测细胞凋亡。与空白组相比较,试验组小肠黏膜上皮细胞的Na+-K+-ATP酶活性降低(P<0.05),Ca2+-Mg2+-ATP酶活性降低(P>0.05),细胞增殖能力降低,S期细胞比例降低,细胞凋亡明显。猪传染性胃肠炎病毒可以降低猪小肠黏膜上皮细胞的Na+-K+-ATP酶活性、Ca2+-Mg2+-ATP酶活性,减弱细胞的增殖能力并引起细胞凋亡。
To observe the influence of the Transmissible gastroenteritis virus(TGEV) on activities of Na+-K+-ATPase,Ca2+-Mg2+-ATPase,proliferation and apoptosis of swine intestinal epithelial cells(IEC).The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase of IEC were measured,which had been infected by TGEV for 24,48 and 72 h,cell proliferation was evaluated with MTT reduction conversion assay,the flow cytometic detected the cell cycle and apoptosis with PI staining.Compared with control group,activities of Na+-K+-ATPase of IEC were lower(P0.05);and activities of Ca2+-Mg2+-ATPase of IEC were lower(P0.05),the proliferation was inhibited,cells in S phase were decreased and cell apoptosis was increased.TGEV depresses the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase,decreases the cell proliferation and causes the apoptosis of IEC.
