摘要
目的评价MTT甲萘醌增强法在眼科丝状真菌药物敏感性试验中的应用。方法用MTT甲萘醌增强法测定二性霉素B和酮康唑对4株眼科丝状真菌(茄病镰刀菌、串珠镰刀菌、烟曲霉菌和黄曲霉菌)的最低抑菌浓度(MIC)并与常量液体培养基稀释法(MaAFST)进行比较。结果在丝状真菌的药敏试验中,甲萘醌能明显增强MTT的显色反应(镰刀菌加甲萘醌0.1mmol/L,曲霉菌加1mmol/L)。二性霉素B和酮康唑对丝状真菌有明显抑制作用,MTT甲萘醌法所测定的A值,随药物浓度的增加而显著降低,按Jahn氏等标准所测定的两种药物MIC值与MaAFST测定的MIC值接近。结论MTT甲萘醌增强法在丝状真菌药物敏感试验中具有方便、快捷、灵敏的特点,值得进一步研究推广。
ObjectiveToevaluatemenadioneaugmentedMTTassayintheapplicationofantifungalsusceptibilityofocularfilamentousfungi.MethodsTheminimuminhibitoryconcentration(MIC)ofamphotericinBandketoconazoleto4strainsofocularclinicalisolatedfilamentousfungi(Aspergillusfumigatus,Aspergillusflavus,FusariumsolaniandFusariummoniliforme)weredeterminedbymenadioneaugmentedMTTassay.TheseMICvalueswerecomparedwithmacrodilutionantifungalsusceptibilitytest(MaAFST).ResultsThecolorationalreactionofMTTwasenhancedsignificantlybymenadione(0.11mmol/L)inthesusceptibilitytestingoffilamentousfungi.TheresultsshowedthatthefilamentousfungiwasinhibitedsignificantlyinthepresenceofamphotericinBorketoconazole,theabsorbency(A)valuesofmenadioneaugmentedMTTassaywasdecreasedfollowingtheincreaseoftheconcentrationofantifungaldrug.TheMICofamphotericinBandketoconazoleweredeterminedbymenadioneaugmentedMTTassay,correlatedexcellentlywithMaAFST.ConclusionThemenadioneaugmentedMTTassayisconvenientandeffectiveintheevaluationofantifungaleffectinvitro.
