摘要
目的筛选聚维酮碘原位凝胶的处方与制备工艺,为我军提供一种新型战伤急救敷料。方法测定不同浓度泊洛沙姆溶液的初始胶凝化温度和黏度,找到适宜浓度的泊洛沙姆和壳聚糖溶液作为原位凝胶基质,添加聚维酮碘作为抑菌剂。以市售抑菌剂作对照,进行原位凝胶的体外抑菌试验。建立皮肤创伤大鼠模型,评价聚维酮碘原位凝胶的创面修复效果。结果 25%(W/V)泊洛沙姆初始胶凝化温度为33℃,黏度为0.098 Pa.S,适于用作战伤急救原位凝胶基质。聚维酮碘原位凝胶抑菌圈(5.3 cm)明显大于市售邦尔康抑菌霜抑菌圈(4.3 cm),抑菌效果明显。动物模型研究证明聚维酮碘原位凝胶能有效预防感染、促进伤口愈合。结论聚维酮碘原位凝胶作为战伤急救敷料具有抗感染、促伤口愈合功能,可用于战创伤尤其是烧伤治疗。
Objective To obtain the optimal formulation of povidone iodine in-situ gel,and to provide a new wound-healing dressing for the military.Methods An optimal formulation of in-situ gel substrate containing poloxamer,chitosan and povidone iodine was selected by measuring the incipient gelation temperature and viscosity of poloxamer at different concentrations.The anti-bacterial effect of the gel was compared with marketed products.Wound healing of the gel was evaluated based on the rat model of skin wound.Results Incipient gelation temperature and viscosity of 25%(W/V) poloxamer solution were 33℃ and 0.098 Pa·S,respectively.Therefore,it could be used as a substrate of first-aid wound healing dress.The inhibition zone of povidone iodine in-situ gel was 5.3 cm in diameter,obviously larger than that of the marketed bacteriostatic(4.3 cm).The gel could prevent infection and improve wound healing according to the result of the animal model.Conclusion Povidone iodine in-situ gel as wound healing dress is effective for anti-infection and wound healing,applicable to the treatment of battle injury,especially burning.
