摘要
目的:研究米曲霉发酵玉米秸秆产富含纤维素酶的饲料的最优条件。方法:采用响应面法对发酵条件进行了优化。对Plackett-Burman设计筛选出的麸皮与秸秆比值、固液比和发酵时间三个主要因素再利用Box-Behnken设计进行优化。结果:确定了以上三个因素的最佳值分别为秸秆:麸皮比值为1.32,固液比为0.68,发酵时间为6.1d。在优化的培养基中,纤维素酶活力为522.36U/g,比优化前的469.13U/g高了11.35%。结论:利用响应面法获得了米曲霉发酵玉米秸秆产纤维素酶饲料的最佳发酵条件。
Objective:The condition of corn stalk fermented by Aspergillus oryzae to produce feed with much cellulase was optimized with response surface methodology(RSM).Method:The major factors of bran to stalk ratio,solid to liquid ratio and fermentation time were screened with Plackett-Burman experimental design.Then the best compounding of fermentation condition was confirm after above major factors were optimized with Box-Behnken experimental design.Result:When bran to stalk ratio was 1.32,solid to liquid ratio was 0.68 and fermentation time was 6.1 day,theoretical value of acid protease was 522.36U/g,which was higher by 11.35% than 469.13U/g of initial medium.Conclusion:The optimal fermentation conduction was obtained for producing feed with much cellulose by Aspergillus oryzae with response surface methodology.
