摘要
采用紫外在线检测方法,对豆豉纤溶酶产生菌UγD8菌株在不同氮源下连续发酵56h,对其发酵特性进行研究,研究结果表明,以酵母膏为氮源发酵产酶时间44h,吸光度Abs最大,为1.4002;以豆浆为氮源发酵产酶时间为36h,Abs为1.0557;以豆渣为氮源发酵产酶时间为48h,Abs为1.3301。所用枯草杆菌UγD8发酵生产豆豉纤溶酶的最佳氮源为酵母膏,产酶量大,但产酶时间晚。其利用率顺序为酵母膏>豆渣>豆浆。选用紫外吸收光谱技术用于检测发酵液中有机氮源消耗以及豆豉纤溶酶的产生等动态变化,得到较为理想的结果,且方便快捷,灵敏度高,可以广泛用于发酵过程的监测。
UV line detection method was utilized in this experiment, strain UyD8 with high productive fibrinolytic enzyme fermented for 56h under various nitrogen sources,the fermentation liquid was detected by UV every 4h. Conclusions showed as below: when yeast extract as nitrogen source, after fermentation for 8h,the new material began to produce, after fermentation for 44h, maximum absorbance (1.4002) appeared, fermentation end, the absorbance decreased.When soybean milk was the sole nitrogen source,the time of new material started to produce was at 4h fermented,in the 36h, absorbance value achieved maximum ( 1.0557).Soybean dreg as nitrogen sources, when fermented for 4h,the broth generated new material,in the 48h,the absorbance value climaxed at 1.3301.
