摘要
研究了澳蜡花‘Snow Flake’组织培养快速繁殖中,外植体消毒的影响因素,旨在得出最佳的消毒方法。试验分别从灭菌剂及灭菌时间,外植体取材时间、取材部位以及抗菌素几个方面进行研究。结果表明:在10月到翌年3月取材,以顶芽为外植体,在75%的酒精中浸泡30s后用0.1%HgCl2处理8min,消毒效果较好。而在培养基中添加抗菌素,对‘Snow Flake’内生菌污染控制效果不理想。由此可见,澳蜡花‘SnowFlake’外植体消毒与灭菌剂及灭菌时间及其取材时间、取材部位相关。
The effect of explant sterilization of Chamelaucium uncinatum＇ Snow Flake ＇ in tissue culture was studied,such as disinfectors,the time of fetching material,cutting sites and antibiotics,to obtain the optimum method of explant sterilization.Result showed that,the optimum explants were terminal bud cut from October to March in next year,and the optimum way of sterilizing treatment was that explants were dipped in 75% alcohol for 30s,then in 0.1% HgCl 2 for 8 min.However antibiotics added into media can not inhibit contamination caused by endophytes.Therefore,explant sterilization of Chamelaucium uncinatum＇ Snow Flake ＇ is related to disinfectors,the time of fetching material and cutting sites.
