摘要
目的端粒酶是迄今为止发现的最为广谱的肿瘤标志物,端粒酶RNA的反义寡核苷酸能够抑制由A549细胞株构建的裸鼠抑制瘤的生长,本研究旨在讨论其机制。方法利用笔者前期实验获得的裸鼠抑制瘤组织,用流式细胞术检测反义寡核苷酸组(antisense oligodeoxynucleotide group,ASODN)、正义寡核苷酸组(sense oligodeoxynucleotide group,SODN)和生理盐水组(normal saline group,NS)移植瘤组织中癌细胞的凋亡指数。结果反义寡核苷酸组、正义寡核苷酸组和生理盐水组的凋亡指数分别是(23.01±2.98)%、(5.48±1.20)%和(3.11±0.93)%,反义寡核苷酸组与正义寡核苷酸组之间、反义寡核苷酸组与生理盐水组之间的差异均有显著性(P值均小于0.01),正义寡核苷酸组与生理盐水组之间的差异无显著性(P=0.056)。结论端粒酶RNA的反义寡核苷酸抑制裸鼠移植瘤生长的机制可能是通过抑制端粒酶活性后诱导癌细胞发生了凋亡。
【Objective】 Telomerase has been the most broadspecum tumor marker so far and antisense oligodeoxynucleotide targeting telomerase RNA can inhibit growth of implanted tumors in nude mice,here,we implore the mechanism. 【Methods】The apoptosis index growth of implanted tumor in Group ASODN (antisense oligodeoxynucleotide group),Group SODN (sense oligodeoxynucleotide group) and Group NS (normal saline group) were detected by flow cytometry.【Results】The apoptosis index of Group ASODN,Group SODN and Group NS was (23.01±2.98)%,(5.48±1.20)% and (3.11±0.93)%,respectively. The difference between Group ASODN and Group SODN was significant in statistics (P〈0.01),and the difference between Group ASODN and Group NS was also significant in statistics (P〈0.01),while the difference between Group SODN and Group NS was not significant in statistics (P =0.056). 【Conclusion】 The mechanism that represses the tumor growth may be that the antisense oligodeoxynucleotide induces cancer cell to apoptosis by inhibiting telomerase activity.
