摘要
本文用14种识别6碱基的限制性内切酶ApaⅠ、BamHⅠ、BglⅠ、BglⅡ、DraⅠ、EcoRⅠ、EcoRⅤ、KpnⅠ、PvuⅡ、PstⅠ、SacⅠ、SalⅠ、SmaⅠ和XhoⅠ研究了来自我国12个省和自治区共计18个地方山羊品种218个个体mtDNA的RFLP,并运用Nei氏公式计算了各限制性类型间的遗传距离P和群体遗传多态度π值。结果表明,在研究的所有个体中共检测到41个酶切位点,18种限制性态型,其中BamHⅠ、BglⅡ、DraⅠ、EcoRⅠ和SalⅠ共5种酶表现出多态。18种限制性态型可归结为6种基因单倍型,单倍型Ⅰ(BamHⅠB、BglⅡA、DraⅠA、EcoRⅠA和SalⅠA)和单倍型Ⅱ(BamHⅠB、BglⅡA、DraⅠA、EcoRⅠA和SalⅠB)为两种基本单倍型,研究结果提示我国地方山羊品种起源于两种不同的母系祖先。各限制性类型间的平均遗传距离为000436,整个群体的平均遗传多态度π值为00487%,表明我国地方山羊品种mtDNA遗传多样性比较贫乏,分化程度较低。
Fourteen restriction endonucleases recognizing 6 base pair,Apa Ⅰ,BamH Ⅰ,Bgl Ⅰ,Bgl Ⅱ,Dra Ⅰ,EcoR Ⅰ,EcoR Ⅴ,Kpn Ⅰ,Pst Ⅰ,Pvu Ⅱ,Sac Ⅰ,Sal I,Sma Ⅰ and Xho Ⅰ were used to investigate the mitochondrial DNA(mtDNA)diversity of 218 individuals of 18 native goat breeds from 12 provinces and autonomous region in China.The genetic distance between haplotypes and the polymorphic degree(π value)were also calculated according to the methods of Neis.The results indicated that 5 restriction endonucleases,BamH Ⅰ,Bgl Ⅱ,Dra Ⅰ,EcoR Ⅰ and Sal Ⅰ,were polymorphic,and 41 restriction sites and 18 restriction morphs which could be sorted into 6 haplotypes were detected.It was shown that haplotype Ⅰ(BamH Ⅰ B,Bgl Ⅱ A,Dra Ⅰ A,EcoR Ⅰ A and Sal Ⅰ A)and Ⅱ(BamH Ⅰ B,Bgl Ⅱ A,Dra Ⅰ A,EcoR Ⅰ A and Sal Ⅰ B)were two main haplotypes.It was inferred that the native goat breeds in China might origin from two different maternal ancestors.The average genetic distance between haplotypes and the polymorphic degree of populations were 0 00436 and 0 0487% respectively.It was also inferred that the genetic diversity and the differentiation of native goat breeds in China was low.
