摘要
目的探讨灯盏花素对家兔心肌缺血再灌注损伤(MIRI)的保护作用。方法将19只家兔随机分为假手术组(6只)、模型组(7只)、治疗组(6只)。以穿线结扎左冠状动脉前降支制备大鼠心肌缺血/灌注模型,治疗组于结扎前5min于耳缘静脉注射灯盏花素,观察各组心肌超微结构,右室血清乳酸(LA)、游离脂肪酸(NEFA)和心肌组织中LA、NEFA、三磷酸腺苷(ATP)水平的变化。结果与假手术组比较,模型组血清及心肌组织中LA、NEFA水平明显升高(P<0.05或P<0.01),心肌组织中ATP含量显著降低(P<0.01),心肌超微结构损害明显;与模型组比较,治疗组血清及心肌组织中LA、NEFA水平明显降低(P<0.05或P<0.01),心肌组织中ATP含量显著增高(P<0.01),心肌超微结构损害减轻。结论灯盏花素可通过改善心肌能量代谢障碍和保护线粒体结构对MIRI起到保护作用。
Objective To explore the protective effect of Breviscapine on myocardial ischemia reperfusion injury(MIRI) in rabbits and its mechanism.Methods Nineteen rabbits were randomized into three groups:sham operated group (n=6),ischemia/reperfusion model group (n=7),and Breviscapine treatment group (n=6). The ischemia/reperfusion models were prepared with the thread-ligation of left anterior descending coronary artery. Five minutes before the ligation,the treatment group was treated with the ear intravenous injection of Breviscapine. The myocardial ultrastructure of rabbits; changes of serum lactic acid (LA) and non-esterified fatty acid (NEFA) of the ventriculus dexter,and the LA,NEFA and adenosine triphosphate (ATP) in the myocardial tissues were detected.Results Compared with the sham operated group,the content of LA and NEFA in the serum and myocardial tissues in the model group was significantly increased (P〈0.01 or P〈0.05),the content of ATP in the myocardial tissues was significantly decreased (P〈0.01),and the myocardial ultrastructure was significantly injured. Compared with model group,the content of LA and NEFA in the myocardial tissues in the treatment group was significantly decreased (P〈0.01 or P〈0.05),the content of ATP in the myocardial tissues was significantly increased (P〈0.01),and the injury of myocardial ultrastructure was significantly relieved.Conclusion Breviscapine is effective in the protection of myocardial tissues injured by ischemia and reperfusion through improving the cardiac energy metabolism and protecting the structure of mitochondria.
