摘要
目的探索结核分枝杆菌RelE毒素蛋白对动物体内的肺癌A-549细胞的成瘤性的影响,以及因此产生的对动物免疫功能的作用。方法构建BALB/C小鼠的免疫抑制模型,通过接种稳定表达结核分枝杆菌RelE毒素蛋白、RelB抗毒素蛋白和RelBE毒素-抗毒素蛋白的肺癌A-549细胞株,含空质粒PcDNA3.1(+)的A-549细胞株,以及正常的肺癌A-549细胞,构建BALB/C小鼠荷瘤模型,观察各组小鼠的肿瘤生长情况,并测定各组小鼠的脾淋巴细胞增殖活性以及血清IFN-γ和IL-2的浓度。结果接种了表达RelE毒素蛋白的A-549细胞的小鼠组,肿瘤生长较其它实验组慢,肿瘤结节的瘤体体积和数量都较其它实验组有明显差异(P<0.05)。对各组小鼠免疫功能变化的研究发现,各实验组小鼠的脾淋巴细胞增殖活性以及血清IFN-γ和IL-2的浓度的差异没有统计学意义(P>0.05)。结论结核分枝杆菌RelE毒素蛋白对肺癌A-549细胞的成瘤性有抑制作用。
Objective To investigate the effect of RelE toxin protein of Mycobacterium Tuberculosis on lung cancer A-549 cell.Methods The immunosuppression BALB/C mice were induced by intraperitoneal injection of CP.The mice were divided into five groups and seeded with the A-549 cells,A-549 cells that stable express RelE toxin protein,RelB antitoxin protein,RelBE toxin-antitoxin protein of mycobacterium tuberculosis and the A-549 cells contained empty vector PcDNA3.1(+).The tumor growth,spleen lymphocyte proliferation and serum IFN-γ and IL-2 were observed.Result The tumor growth of the mice seeded with A-549 cells contained RelE toxin protein was slower than that of the other groups.The number of tumor nodus of the mice seeded with A-549 cells contained RelE toxin protein was less than that of the other groups.Conclusion RelE toxin protein of mycobacterium tuberculosis has tumorigenicity inhibition to lung cancer A-549 cell.
