摘要
目的观察健儿强身膏对哮喘豚鼠血清IL-10、IL-6、TNF-α表达的影响。方法采用卵蛋白(OVA)致敏法制备豚鼠哮喘模型,将48只豚鼠随机分为6组,即空白对照组、模型组、阳性对照组、健儿强身膏(高、中、低)3个剂量组,观察各组引喘潜伏期(T)及肺组织的病理变化,应用酶联免疫吸附试验法测定血清IL-10、IL-6、TNF-α的表达水平。结果各治疗组均可延长引喘潜伏期,与模型组相比差异有统计学意义(P<0.05)。与空白对照组相比,模型组的IL-6、IL-10表达显著增加(P<0.01),TNF-α的表达无显著差异;各治疗组与模型组相比IL-6、IL-10表达显著下降(P<0.01),TNF-α的表达无显著差异。结论健儿强身膏能下调哮喘豚鼠IL-6、IL-10的表达,可能为其抑制哮喘呼吸道炎症的重要作用机制之一。
Objective To observe the effect of Jianerqiangshen electuary on expression of IL-10,IL-6 and TNF-α on asthmatic guinea pigs.Methods The asthma guinea pig model was established by the ovalbumin(OVA) challenge methods.Forty-eight guinea pigs were randomly allocated to six groups:the blank control group,asthma model group,dexamethasone-treated group,three dose groups of Jianerqiangshen electuary.The effects of relieving asthmatic attacking were observed and the morphology structures were observed.The concentrations of IL-10,IL-6 and TNF-α were measured by sandwich enzyme-linked immunosorbent assay.Results Compared with the normal group,IL-6 and IL-10 of the model group significantly increase(P 〈0.05).The IL-6 and IL-10 in guinea pigs were obviously lower than that of the asthmatic guinea pigs(P 〈0.01) after Jianerqiangshen electuary treatment,and TNF-α was not different between the groups treated respectively and the model groups.Conclusion Jianerqiangshen electuary can depress IL-6 and IL-10 expression,and it may be a key process in modulatory mechanism of asthma.
