纤维蛋白凝胶携载血管内皮生长因子对大鼠损伤坐骨神经血管新生的影响 被引量:1

Effect of fibrin glue carried with exogenous vascular endothelial growth factor on angiogenesis in rats after sciatic nerve injury

摘要 目的探讨局部应用纤维蛋白凝胶携载血管内皮生长因子对损伤坐骨神经组织血管生成的影响。方法将36只Wistar大鼠左侧坐骨神经切断,神经两断端原位缝合,制作大鼠坐骨神经损伤动物模型。然后将大鼠随机分成实验组(纤维蛋白凝胶携载血管内皮生长因子给药组)与对照组(血管内皮生长因子基因转染给药组),每组18只,于用药后4、8、12周行肉眼观察、神经微血管计数检查。结果实验组用药后4、8周的神经微血管计数显著低于对照组(P<0.05),用药后12周两组间差异无统计学意义(P>0.05)。结论纤维蛋白凝胶可以作为血管内皮生长因子的载体,纤维蛋白凝胶携载血管内皮生长因子给药可以促进神经血管新生。 Objective To investigate the effects of fibrin glue(FG)carried with exogenous vascular endothelial growth factor(VEGF)on angiogenesis in rats after sciatic nerve injury.Methods The rat sciatic nerve injury model was made by cutting off the left sciatic nerve of rat and suturing in normal position.Thirty-six Wistar rats were randomized into 2 groups:experimental group(FG carried with VEGF,n=18)and control group(VEGF gene transfection,n=18).The gross appearance was observed and microvascular count(MVC)was calculated at weeks 4,8,12 after medication.Results There was statistical difference in MVC of the nerve between 2 groups at weeks 4,8(P〈0.05)after medication,but not at week 12(P〉0.05).Conclusion Fibrin glue can be used as a carrier for VEGF to repair the injured sciatic nerves.The results indicate that fibin glue carried with VEGF can exert the angiogenesis in the injured sciatic nerve area of rats.
