大鼠活体心脏穿刺采血法 被引量:6

A Method of Gathering Blood by Intracardiac Penetration in Living Rats

摘要 报告一种大鼠活体多次采血法。用1 % 戊巴比妥钠腹腔麻醉后,剑突下进针,斜行向上刺入2-5 ~3 .0cm 即达心脏,回抽。30 只SD大鼠,间隔10 d 采血2 ml,共4 次,测定Hb、RBC、Hct、WBC、ALT、TP、BUN。结果:采血1 000 余次,1 次穿刺成功率98 % 以上,且大鼠成活,各次血液成份及生化指标差异均无显著性( P> 0-05) 。本法操作简便,定位准确,采血量大,可重复采血。 We have established a new method which can gather more blood repeatedly in living rats.The rats were anaesthetized by intraperitoneal injection with 1% sodium pentobarbital,then penetrated upward obliquely at the site of just below the xiphoid,entered about 2.5~3.0 cm into the heart and drew blood out.Two ml blood has been extracted from 30 SD rats every 10 days 4 times in all,and the Hb,RBC,Hct,WBC,ALT,TP and BUN have been determined.Results:by using this method,the rate of one time successful penetration is over 98% without sacrificing animals,and no significant difference ( P >0.05) has been detected in blood components and biochemical targets each time.The method is easy to control and is applicable for gathering more blood repeatedly in living rats.
