摘要
目的:探讨激活素A对原代培养小鼠腹腔巨噬细胞活化的影响。方法:ELISA法检测激活素A,RT-PCR法检测iNOS mRNA表达,流式细胞术检测CD86及Arginase-1的表达。结果:LPS以时间依赖方式促进小鼠腹腔巨噬细胞分泌激活素A,激活素A能明显促进巨噬细胞活化标志CD86分子表达。LPS明显促进Ⅰ型巨噬细胞标志iNOS mRNA表达,而激活素A则主要促进Ⅱ型巨噬细胞标志Arginase-1的表达。结论:激活素A可能以自分泌/旁分泌形式促进小鼠II型巨噬细胞活化。
Objective:To investigate the effect of activin A on the activation of mouse peritoneal macrophages.Methods:Activin A level was measured by ELISA,iNOS mRNA expression was detected by RT-PCR,and expressions of CD86 and arginase-1 was examined by flow cytometry.Results:LPS promoted secretion of activin A by the peritoneal macrophages of mice in a time-dependent manner.Activin A enhanced the expression of activation marker CD86 on the surface of macrophages.Further more,the results showed that LPS promoted the expression of iNOS mRNA,the M1 marker,whereas activin A mainly induced M2 marker,arginase-1 expression in mouse peritoneal macrophages.Conclusion:Activin A may mainly induce M2 activation by autocrine/paracrine manners.
