摘要
以姜荷花新品种"红观音"球茎为外植体进行组织培养与快速繁殖研究。结果表明:外植体经灭菌处理后接种到MS+BA2~3mg/L+NAA0.2~0.3mg/L培养基上,均能较好地诱导休眠芽的生长;丛生芽在18号培养基(花宝2号1.5g/L+MgSO40.15g/L附加MS铁盐及其维生素和其它有机物+椰子汁100mL/L)+TDZ0.5mg/L培养基上增殖效果最佳,增殖系数可达3.73,且玻璃芽少;生根培养基以18号培养基+NAA0.5mg/L效果最好,生根率为92%;试管苗在以园土:泥炭土:珍珠岩=1:1:1(V/V/V)的栽培基质中,成活率可达95%以上。
Tissue culture and rapid propagation of Curcuma alismatifoliacv.Red Guanyin were studied.When sterilized corms that were cut into piece with 1-2dormancy buds were cultured on MS medium with 2-3mg/L 6-BA and 0.2-0.3mg/L NAA,budding and cluster buds could be induced.The proliferation rates were 3.73in H18(Haponex 1.5g/L and MgSO40.15g/L instead of macro-element and micro-element of MS)+coconut milk 100mL/L+TDZ 0.5mg/L.The best medium for rooting was H18contained 0.5mg/L NAA and the rooting rate was 91.43%.When the test-tube plantlets were transplanted on humus:sphagnum:perlite=1:1:1(V/V/V),their survival rate of was over 95%.
