摘要
目的:研究何首乌不同炮制品对大鼠肝细胞色素P4502E1(CYP2E1)基因mRNA表达的影响。方法:50只大鼠随机分为空白组、清蒸组、黑豆汁制首乌A、B、C 5组,给药组每日灌胃主药5g.kg-1,空白组连续给同等体积生理盐水,连续给药90d,实验结束时采用逆转录聚合酶链反应(RT-PCR)法测定大鼠肝CYP2E1基因mRNA在给药后的变化。结果:各给药组CYP2E1基因mRNA表达水平明显低于空白组(P<0.01),各给药组组间差异无显著性(P>0.05)。结论:首乌不同炮制品均能够显著抑制CYP2E1mRNA的表达。
OBJECTIVE To study the effect of different Radix Polygoni multiflori preparata on the mRNA expression of CYP2E1 gene of the liver in rats. METHODS 50 rats were randomly divided into the control group, the steamed group, and Polygonum muhiflorum Thunb （A,B, C） 5 groups, each comprising10. The administration groups were given orally 10 g·kg-1 each day, and the control group received isovolumic saline by gavage. The mRNA levers of the expression of the CYP2E1 gene of the rat liver were assayed with RT-PCR after the drug administered orally for 9{） d. RESULTS Each of the CYP2EI mRNA level of the administration groups was significantly lower than control group （P〈0. 01 ）and there was no marked difference for each of administration group（P〉0. 05） . CONCLUSION The different Radix Polygoni multiflori preparata can sig- nificantly inhibit the expression of CYP2E1 mRNA.
