摘要
[目的]研究黄粉虫不同发育时期酚氧化酶(PO)的活性及其酶原(PPO)mRNA表达量差异。[方法]用3种不同的处理方式处理黄粉虫末龄幼虫、蛹和成虫。24 h后测定血淋巴的PO活性,并通过半定量RT-PCR方法检测PPO的表达量。[结果]PO活性表现出随龄期推进活性加强的趋势,即成虫的活性最高,蛹次之,末龄幼虫最低。没有经过处理时,蛹期几乎没有PPO基因的表达。经细菌处理后,PPO表达量增加,但末龄幼虫最低。[结论]黄粉虫不同发育时期的血淋巴PO活性以及表达量存在差异,具有发育时期相关性。
[Objective] The aim was to investigate the differences of PO activity and the mRNA expression during different developmental stages of Tenebrio molitor.[Method] Tenebrio molitor were chosen from each developmental stage(last instar larvae,pupae and adults) to be treated with three methods.The PO activity was measured and PPO expression was detected by semi-quantitative RT-PCR after 24h.[Result] PO activity increased with ontogeny,which was most vigorous in adults,next in pupae and then in last instar larvae.Negligible expression of PPO mRNA in control group from pupae,but increased significantly after bacteria challenge and was lowest in last instar larvae.[Conclusion] There is a difference of PO activity and the mRNA expression in haemolymph of Tenebrio molitor during different developmental stages and it is developmental stages-related.
