XRCC1、DNA-PKcs和PCNA在非小细胞肺癌中的表达及其与放疗疗效的关系研究 被引量:2

摘要 目的探讨X线修复交叉互补基因(XRCC1)、DNA-PKcs和增殖细胞核抗原(PCNA)在非小细胞肺癌(NSCLC)中的表达及其与放疗疗效的相关性,为预测NSCLC放疗的敏感性提供理论依据。方法 62例经病理活检证实的NSCLC患者行三维适形放射治疗,放疗前后胸部增强CT扫描,按照WHO实体瘤近期疗效标准进行评价;用免疫组化染色法检测放疗前NSCLC组织中XRCC1、DNA-PKcs和PCNA蛋白的表达情况;将放疗的近期疗效与3种蛋白免疫组化结果进行统计分析。结果按XRCC1蛋白(-)、(+)、(++)、(+++)分组经放疗后有效率(CR+PR)分别为70.0%、66.7%、64.0%、40.0%,组间比较差异有统计学意义(P<0.05);按DNA-PKcs蛋白(-)、(+)、(++)、(+++)分组经放疗后有效率分别为85.7%、76.5%、55.6%、27.3%,组间比较差异有统计学意义(P<0.05);按PCNA蛋白(-)、(+)、(++)、(+++)分组经放疗后有效率分别为0、38.5%、60%、70.8%,组间比较差异有统计学意义(P<0.05)。结论 XRCC1、DNA-PKcs蛋白表达与NSCLC三维适形放射治疗的近期疗效呈负相关,PCNA蛋白表达与其呈正相关,检测这3种蛋白的表达在一定程度上可作为预测NSCLC三维适形放射治疗的近期疗效指标。 Objective To investigate expressions of XRCC1,DNA-PKcs and PCNA proteins in non-small cell lung cancer(NSCLC)and its relationship to the effect of radiotherapy.Furthermore,to analyze the predict value of radiotherapy effect.Methods There were 62 patients with NSCLC who were treated by 3D-radiotherapy.Before and after the radiotherpy,they were checked by chest enhanced-CT to evaluate the short-term effect.The expression of XRCC1,DNA-PKcs and PCNA in 62 patients with NSCLC were determined by immunohistochemical method.The expression levels of the three kinds of proteins with the short-term radiotherapy effect were analyzed.Results The expressions of XRCC1,DNA-PKcs and PCNA were closely related with short-term effect after radiotherapy in NSCLC.The short-term effect was descent along with enhancement of XRCC1 and DNA-PKcs,and was rised along with enhancement of PCNA.Conclusion These findings indicated that the expressions of XRCC1,DNA-PKcs and PCNA may predict short-term effect of radiotherapy.The three proteins may have relationship with radiotherapeutic sensitivity in some extent.
