MIRU-VNTR技术对源于河南的北京基因型结核分枝杆菌分型研究 被引量:13

摘要 目的:研究MIRU-VNTR技术对结核分枝杆菌北京基因型家族的分辨能力。方法:用检测RD105区缺失的多重PCR(DTM-PCR)方法对河南省源的结核分枝杆菌进行北京基因型鉴定,鉴定后的菌株检测基因分型方法VNTR-7和VNTR-16位点的分辨能力。结果:DTM-PCR获得98株结核分枝杆菌北京基因型家族菌株,VNTR-16和VNTR-7分辨率指数(Hunter-Gaston指数,HGI)为0.9983、0.9953,菌株的成簇率分别为3.1%、11.2%。结论:VNTR-16和VN-TR-7位点基因分型方法用于北京基因型家族的分析有较高的分辨能力,操作简便,适于结核病控制工作中北京基因型家族菌株占优势地区的分子流行病学研究。 Objective:To explore the power of discriminating the Beijing family geno type strains of Mycobacterium tuberculosis using MIRU-VNTR detection method.Methods:To distinguish the Beijing family genotype isolates with the non-Beijing family genotype using deletion-targeted multiplex PCR(DTM-PCR) method,than the Beijing family isolates were used to estimate the discriminate power of VNTR-16 loci set and VNTR-7 loci set detection method.Results:98 Mycobacterium tuberculosis strains of Beijing genotype family were obtained with DTM-PCR,the HGI value of VNTR-16 and VNTR-7 is 0.9983 and 0.9953,the percentage clustering of VNTR-16 and VNTR-7 is 3.1% and 11.2%.Conclusion:The methods of VNTR-16 and VNTR-7 both show high discrimination power and easy operation for distinguishing the Beijing family genotype strains of Mycobacterium tuberculosis and they are suitable for application to TB control program where the Beijing family genotype strains predominate.
