摘要
为了在Marc-145细胞上获得更高滴度的猪繁殖与呼吸综合征病毒(PRRSV)NVDC-JXA1株,优化了细胞培养条件、病毒接种剂量、病毒吸附时间、收毒时间和病毒冻融次数等条件。结果表明Marc-145细胞在MEM培养基、新生牛血清含量80 mL/L,细胞接种密度150 000个/mL,pH7.0条件下生长状态最好;PRRSV NVDC-JXA1株的最佳接种剂量为400 TCID50/mL,病毒吸附时间为30 min,收毒时间为接种后72 h,在-20℃冻融3次,PRRSV NVDC-JXA1株增殖效果最好。该结果为NVDC-JXA1株PRRSV疫苗的生产提供了依据。
To obtain higher titers of porcine reproductive and respiratory syndrome virus (PRRSV) NVDC- JXA1 strain, the factors influencing the propagation of PRRSV-MVDC-JXA1 strain on Marc-145 cell, such as medium, cell concentration, pH of the medium, the inoculation dose, the absorbtion time, the harvest time, freeze-thawwed times, were optimized. Results showed that the optimum factors for PRRSV propaga- tion were as follows: the medium was MEM plus 80 mL/L of fetal bovine serum; cell concentration was 150 000/mL; the pH of the medium was 7.0; the inoculation dose of PRRSV was 400 TCID50/mL; the absorbtion time was 30 minutes; the harvest time was 72 hours after virus inoculation; the virus needed to be freeze-thawwed 3 times at --20℃. This study provided a basis for large-scale production of PRRS NVDC-JXA1 strain vaccine.
