摘要
目的:建立人血清中25-羟基维生素D3、维生素A的高效液相色谱测定方法。方法:血清以甲醇沉淀蛋白质,用正己烷提取25-羟基维生素D3、维生素A,吹氮浓缩后用HPLC测定。结果:25-羟基维生素D3、维生素A的线性范围分别为6.7~335.5nmol/L、0.13~6.64μmol/L,方法回收率在90%~110%之间,萃取回收率大于70%,日内日间RSD小于6%。结论:该方法灵敏、准确、快速,其测定的结果可为临床对相关疾病的诊断提供参考依据。
Objective:To establish an HPLC method for the determination of 25-hydroxy-vitamin D 3and vitamin A in human serum.Methods:Proteins were precipitated with methanol.Vitamin A and 25-hydroxy-vitamin D 3in human serum were extracted with n-hexane and determined with HPLC.Results:The linear ranges for the 25-hydroxy-vitamin D 3 and vitamin A were 6.7~335.5 nmol/L and 0.13~6.64μmol/L,respectively.The relative recovery averaged was between 90%~110%,the absolute recovery was more than 70%,the mean inter-and intra-assay coefficients variation was less than 6%.Conclusions:The method is sensitive,accurate and quick.Results of determination can provide a reference for relevant clinical diagnosis of the disease.
