摘要
目的:观察淫羊藿乙酸乙酯提取物和正丁醇提取物对D-半乳糖致衰老小鼠行为学和脏器指数的影响,探讨其抗衰老作用和功能药性部位。方法:采用D-半乳糖致衰老小鼠模型,分别灌胃给予53mg/kg、169mg/kg的淫羊藿乙酸乙酯提取物和85mg/kg、272mg/kg的淫羊藿正丁醇提取物,测试小鼠抓力、自主活动、悬尾及水迷宫的学习记忆成绩,并观察对脏器指数、骨骼重量和强度的影响。结果:淫羊藿乙酸乙酯提取物、正丁醇提取物均可明显缩短水迷宫测试到达平台的时间和游泳总路程,对抓力、悬尾实验、骨骼重量和强度无明显影响;淫羊藿正丁醇提取物低剂量组可减少小鼠的自主活动并增加睾丸指数。结论:淫羊藿乙酸乙酯提取物和正丁醇提取物能改善D-半乳糖致衰老小鼠的空间学习记忆能力,淫羊藿正丁醇提取物可增加睾丸指数。
Objective:To investigate the anti-aging effects and functional herbal nature site of Epimedium with different extracts on D-galactose (D-gal)-induced aging mice. Methods:The low-dose(53 mg/kg)and high-dose (169 mg/kg)of Epimedium ethyl acetate,low-dose(85 mg/kg)and high-dose (272 mg/kg)n-butanol extract were respectively given to the aging model mice by intragastric gavage. Then the behavioral characteristics,ability of learning and memory,organ index and effect on the bone were measured. Results:Epimedium ethyl acetate and n-butanol extract shorted the time for reaching platform and swimming distance in Morris Water Maze test (P0.01,P0.05). Meanwhile,the grip strength test,tail suspension test,weight and strength of bone were not different from those of control group (P0.05). The low-dose of Epimedium n-butanol extract decreased the number of spontaneous activities and improved the testis index significantly (P0.05). Conclusions:Epimedium ethyl acetate and n-butanol extract can improve the ability of learning and memory in aging model mice. Epimedium n-butanol extract can increase the testis index which may be related to the effect on growth of reproductive organs.
