摘要
目的:凝胶渗透色谱法测定胶原肽分子质量及其分布。方法:采用三根TSK gel G2000 SWXL(7.8 mm×30 cm,Tosoh)色谱柱串联,流动相为乙腈-水-三氟乙酸溶液(10:90:0.1,v/v/v);流速:0.8 ml/min;检测波长:220 nm;柱温:30℃。结果:在分子质量149 Da~13700 Da之间建立了标准曲线lgMr=-0.126t+7.228,相关系数r=0.994,方法精密度(RSD)小于5.97%。结论:应用本法测定胶原肽分子质量及其分布具有操作简单、精密度高、结果可靠、易于推广的优点。
Objective:To establish a method of gel permeation chromatography to determine molecular weight and its distribution of collagen peptide.Methods:Three TSK gel G2000 SWXL(7.8 mm×30 cm,Tosoh) columns in series was equilibrated with mobile phase composed of acetonitrile/water/trifluoroacetic acid(10:90:0.1,v/v/v) at a flow rate of 0.8 ml/min,detection was performed at 220 nm,and the column temperature was set as 30℃.Results:The relationship curve(lgMr=-0.126t+7.228) between the logarithm of molecular weight and retention time was obtained in the molecular weight range of 149 Da to 13700 Da.The relative standard deviation(RSD) of the method were less than 5.97%.Conclusion:The method has advantages of simplicity,preciseness,accuracy and easiness.
