摘要
对天女木兰(M.sieboldiiK.Koch)种子进行变温层积处理,测定其在GA3处理诱导下种子内淀粉、糖含量以及糖代谢关键酶的活性。结果表明:在GA3诱导下,淀粉作为一种暂时储存的物质不断地降解,层积结束后CK处理淀粉含量最少,降至0.43mg·g-1;层积前期(40d)与层积后期(120d)以积累蔗糖为主,以层积40d时CK处理蔗糖含量最大,其值高达2.3mg·g-1,同期酸性转化酶(AI)和中性转化酶(NI)活性降低,蔗糖合成酶(SS)和蔗糖磷酸化酶(SPS)活性升高;层积中期(90d)果糖和葡糖糖含量随GA3浓度的增加有增强的趋势,以1500mg·L-1 GA3处理果糖和葡萄糖含量最高,分别为0.33mg·g-1和0.38mg·g-1,此时AI和NI活性达到最大值。
In order to understand the characterristics of suger metabolism in M.sieboldiiK.Koch seeds,monitored the changes of starch,suger and sucrose metabolism enzyme activities were monitored on GA3 induced of M.sieboldiiK.Koch seeds during variable-temperature stratification process.The results showed that the starch as a temporary substance was decomposed continuously on GA3 induced during the stratification process,starch content of CK treatment reduced the minimum value afeter stratification process(0.43 mg·g-1).Sucrose were predominantly accumulated in the early stratification stages(40 days) and late stratification stages(120 days),suger content of CK treatment reduced the maximum value afeter stratification process(2.3mg·g^-1).In the same time period the activities of sucrose synthase(SS) and sucrose phosphate synthase(SPS) rised,while the activities of acid invertase(AI) and neutral invertase(NI) reduced.The sucrose content were decomposed continuously;Glucose and fructose content increased with the increasing amount of GA3,and 1500mg·L^-1 of GA3 treatment had the highest content of glucose and fructose,0.33mg·g^-1 and 0.38mg·g^-1,respectivelies.Meanwhile,the activities of AI and NI reached the maximum.
