热放疗对人肺腺癌耐药细胞株A549/DDP凋亡的影响 被引量:1

摘要 [目的]观察热放疗对人肺腺癌耐药细胞株A549/DDP凋亡作用的影响。[方法]MTT法细胞毒性实验和克隆形成实验检测热放疗对A549/DDP细胞抑制作用和克隆形成的影响;TUNEL法检测热放疗对细胞凋亡作用的影响;透射电镜观察各组细胞形态学改变。[结果]细胞毒性实验和克隆形成实验显示热放疗对A549/DDP细胞生长有明显抑制作用(P<0.01);TUNEL法观察到热放疗有诱导A549/DDP细胞凋亡作用;电子显微镜观察到热放疗后A549/DDP出现细胞核固缩、变形,染色质浓聚、边集等凋亡现象。[结论]热放疗能诱导A549/DDP细胞凋亡。 [Purpose] To investigate the effect of radiohyperthermia on apoptosis in multidrug-resistant human lung adenocarcinoma cell line A549/DDP. [Methods] Effect of radiohyperthermia on A549/DDP cell inhibition ratio and clone formation rate were detected by MTT assay and clone formation experiment. A549/DDP cell apoptosis phenomenon was detected by TUNEL assay. Morphological changes of cells were observed by transmission electron microscopy. [Results] MTT assay and clone formation experiment showed that radiohyperthermia inhibited A549/DDP cell proliferation(P0.01). Radiohyperthermia inducing A549/DDP cell apoptosis was observed by TUNEL assay. Cell apoptostic features such as karyopyknosis, deformation and karyorrhexis, chromatin accumulation were observed by transmission electron microscopy. [Conclusion] Radiohyperthermia could induce A549/DDP cell apoptosis
