摘要
目的观察二烯丙基二硫(DADS)对人结肠癌SW 480细胞增殖的抑制作用。方法实验分DADS处理组和未处理组。MTT法、集落形成实验检测细胞增殖活性,光镜和电镜观察细胞形态学变化,流式细胞术检测周期分布,免疫细胞化学检测增殖细胞核(PCNA)表达改变。结果MTT和集落形成实验显示DADS作用SW 480细胞后,细胞生长抑制率呈浓度、时间依赖性增加,而集落形成率显著降低;组织学显示处理组细胞异型性降低,核质比下降,细胞器丰富;流式细胞仪分析显示DADS明显将细胞周期阻滞于G2/M期,且呈剂量依赖关系;免疫细胞化学显示处理组较未处理组细胞PCNA蛋白表达明显减弱(P<0.05)。结论DADS可体外抑制人结肠癌SW 480细胞增殖,使癌细胞阻滞于G2/M期,可能与下调PCNA蛋白的表达有关。
Objective To investigate the inhibitory effect of diallyl disulfide(DADS) on human colon cancer cell line SW480 in vitro. Methods The experiment was divided into two groups: treated group with DADS and untreated group.SW480 cells proliferation was measured by MTT assay and colony forming.The morphology of tumor was observed under optical microscope and electron microscope.Cell cycle distribution was analyzed by flow cytometry(FCM).The expression of proliferating cell nuclear antigen(PCNA) was detected by immunohistochemistry. Results MTT showed that DADS significantly inhibited SW480 cells proliferation with a dose and time-dependent manner,and the colony-formation rate of SW480 cells treated by DADS significantly decreased with a dose-dependent manner.In treated group,the cellular atypia and nuclear-cytoplasmic ratio were decreased,cell organs were abundant in the intracytoplasm,which were observed by optical microscope and ultramicroscope.Flow cytometry revealed that DADS could markedly arrest cell cycle in G2/M phase with a dose-dependent manner.The expression of PCNA were observed in the two groups by Immunohistochemistry,but it was weakened markedly in the treated group(P〈0.05). Conclusion DADS had significant inhibitory effect on proliferation of human colon cancer SW480 cells in vitro and arrested cell cycle in G2/M,and its molecular mechanism is related to downregulating expression of PCNA.
