摘要 目的寻找神经管畸形新生儿血清中异常表达的蛋白质。方法通过二维凝胶电泳分离神经管畸形新生儿及正常新生儿血清,用PDQuest图像软件比较电泳图谱中的异常蛋白质点,用MALDITOF/TOF串联质谱仪分析蛋白质,所得的质谱图通过GPS软件在Mascot数据库中检索并鉴定蛋白质。结果筛选出35个表达差异的蛋白质点,其中有8个蛋白质点表达上调,27个蛋白质点表达下调。选取4个蛋白质点进行鉴定,其中结合珠蛋白前体表达升高,HP蛋白,血清载脂蛋白A-1前体及免疫球蛋白重链恒定区-1蛋白表达均降低。结论神经管畸形可以导致血清中多种蛋白的的异常表达。 Objective To search for differentially expressed serum proteins in neonates with neural tube defects. Methods Serum proteins from neonates with neural tube defects and the normal babies were separated using two-dimensional (2-D) gel electrophoresis. Differentially proteins spots on the 2-D maps were selected and further analyzed using PDQuest software and matrix-assisted laser desorption/ionization two-stage time of flight (MALDI TOF/TOF) tandem mass spectrometry,respectively,and subsequently matched via general proteomics standard (GPS) in Mascot database. Results Thirty five differentially expressed protein spots were found,among which 4 proteins were evaluated. In neural tube defects neonates the expression of heptoglobin precursor was up-regulated,while HP protein,APOA-1 precursor and IGHG1 protein were down-regulated. Conclusion The neural tube defects can cause differential expressions of many proteins in neonatal serum.
