小鼠血清环磷酰胺浓度HPLC测定方法的建立

摘要 采用岛津高效液相色谱仪,紫外检测器,CBM-102工作站,分析柱为DiamonsilC18(150mm×4.6mm,5m)。血清样品甲醇萃取,风干后,配制成100:l水溶液进样检测。所建立的检测条件为:流动相乙腈-水(18:82 V/V),流速0.5 ml/min;咖啡因为内标,终浓度8mg/ml;检测波长200 nm。结果显示,环磷酰胺标准品线性方程为:线性范围为10-200 mg/l。表明实验中所建立的方法色谱峰分离较好。 In ooder to develop a High Performance Liquid Chromatography (HPLC) method for determining the concentration ofCyclophosphamide (CP) in the plasma of the immunosuppressed mice, the Shimadu HPLC system was used. The analytical column was a diamonsil C 18 column, with the detective UV wavelength of200nm. Acetonitrille and water with the rate of 18 : 82 (V/V) and with the flow rate of 0.5ml/min was used in isocritic mobile phase. The plasma samples were collected, and then were deposited down the protein by methanol. The remains was air-dried and dissolved with 100? 1 water. Caffeine was added as the internal standard with a final concentration of 8mg/ml. The results showed that the liner equation was, It determinated that the chromatography was consistent with the parameters used here.
