摘要
目的观察大黄对慢传输型便秘大鼠结肠传输能力的影响,揭示大黄与该病发展的相关性。方法将健康SD大鼠24只随机分为大黄组及对照组,大黄组应用中药大黄喂养制作慢传输型便秘的动物模型。分别于禁食后,经口灌入活性炭悬液10 mL/kg,动物处死后剖腹取出幽门到直肠末段的全部肠管并测量肠管长度、碳末推进长度及肠道传输功能。取结肠组织进行HE染色及电镜下观察。同时进行免疫组织化学染色,观察血管活性肠肽(VIP)的表达状况。结果大黄组碳末推进长度、肠道传输功能低于对照组。大黄组大鼠结肠肌间神经丛VIP含量较对照组明显下降,光镜及电镜下均可见结肠细胞结构明显退行性改变,结肠肌间神经丛内未找到Cajal间质细胞(ICC)。结论大黄等蒽醌类泻剂损伤结肠壁神经丛,使结肠传输功能下降。因此,对慢传输型便秘患者治疗时,应避免长期应用此类药物。
Objective To observe the influence of Dahuang(Radix et Rhizoma Rhei) on colonic transit in rats with slow transit constipation(STC),and reveal the correlation between Dahuang and STC development.Methods All SD rats(n=24) were randomly divided into the Dahuang group and control group.The rat model of STC was established by feeding Dahuang in the Dahuang group,and after fasting activated charcoal suspension(10 mL/kg) were given orally to rats of two groups respectively.The length of whole intestine,charcoal powder propelling length and colonic transit were detected after killing rats and taking the intestine from pylorus to distal rectum.The colonic tissue was stained by HE and observed with the electron microscope,and at the same time stained by immunohistochemistry for observing the expression of vasoactive intestinal polypeptide(VIP).Results The charcoal powder propelling length and colonic transit were reduced in the Dahuang group compared with those in the control group.The level of VIP in colonic intermuscular nerve plexus decreased significantly in the Dahuang group than that in the control group.The obvious retrograde degenerations of colonic cytoarchitacture were observed by both light microscope and electron microscope.The colonic transit was slower and there was no interstitial Cajal cells (ICC) in colonic intermuscular nerve plexus in the Dahuang group,and VIP level decreased significantly compared with that in the control group.Conclusion Anthraquinone cathartic,including Dahuang,can injure colonic intermuscular nerve plexus and induce colonic transit.This kind of medicines should not be administrated for long time when curing STC.
